I'm looking to upgrade the tires on my stock sr5 16" wheels. Will most likely go with the 265/75r16. I want the wheels to stick out of the fender slightly. Will wheel spacers accomplish this? Do they effect ride or anyting else?

Yes they'll push the wheels out and are recommended when bumping up tire size on stock wheels.

On stock tires 245/?/16 they make it flush with the fender, with 265/75/16 duratracs it will be like 1 or so inches outside of the wheel well. I have spidertrax 1.25 wheel spacers

Depends on how much bigger you're going. Not necessary for 265/75R16 for example, but they can help you clear 285s if you want to stick with stock wheels.

Are used spacers something to be weary of?

i wouldn't worry about buying used spacers. as long as the studs is not rusted out or the spacer itself beat do death, you should be ok. plus you'll be saving about half on a used set.
