So I am trying to get new Sway Bar Bushings for my 1996 Tacoma LX (4wd, 3.4 V6) and I called all my local part stores and no one had them in stock but all of them said they had a few different sizes, one was a 27 and one was like a 24 or 25 I think. Is there a way to check the size or does it really matter?

Your 96 should take the smaller of the 2. The "TRD" package in later years had a 1mm larger diameter bar. IIRC the sizes are 26 and 27mm (bar diameter).

No, you need the smaller bushings mentioned.
