6inch lift sitting on bilstein coilovers. Lexus is300 studs in front to keep stock wheels, general grabber red letters, nfab front bumper.
I'm looking to go up to 37 inch tires on my 2012 4.0 4x4 sometime next year. I want to re gear the rear end but the problem is I have no idea what setup to run with a 37 inch tire and a 17 inch wheel. Also I have the trd locker. Would I have to re gear the front diff as well?

Yes, if you want to use 4wd you will have to regear the front as well.

37 is a big tire. You got a big hammer to go with them?

Supercharger will be the next addition. Along with new suspension from king and alcan leafs and a shackle flip kit.
