After a 10 hour drive home my truck has developed a knock from the rear end. Its most noticeable under 5 mph. Its rhythmic.

I put the truck on stands, put it in neutral and rotated the wheels. I could not replicate it in the garage. I have a feeling its one of the axles or the diff. Any ideas? Any advice on how to diagnose?

Check the level and condition of the oil in the rear axle for a start.

While it is up on the stands inspect the u joints too.

Check your lug nuts. Don't ask me how I know.... just saying.

Does the knock change with speed? Going over bumps? Around corners? Forward vs reverse?

Dumped the Tacoma, that's all it needed.
    Mileage?
    Model?
    Drivetrain options?

if you have a selective locker in the rear, we’re you recently using it?

As already stated, check fluid levels, driveline play, and wheel lugs.

I’d inspect all of the rear suspension too while I was under there just because, and a general condition inspection on everything. And check the pressure in your spare.

    Start easy.

Rotate tires front to back. See if it remains.

Hey guys thanks for all the reponses.
Lugs were the culprit. I had several at less than spec torque. Retighten and the noise is gone.
Thanks again. Love when its simple. And cheap!

Glad it worked out -- for cheap too!
