    I have a 2006 Tacoma 4 wheel drive
Having an issue with the right rear brake locking up.
When I back up then put it in drive and start moving forward
    The brake locks up.
Then while sitting still I can apply the brakes and it will release.
I have changed the wheel cylinder and the drum. And it’s still locking up
Anyone have an idea what could be the problem.

    Welcome

Did you check it out with the wheels off,
drum off and have someone step on the brake
to see how it is operating and problem
    while manually turning the axle.
Safety first, always use jack stands.

I would advise against this as one wrong move and you will blow out your new wheel cyl and contaminate the brake shoes. One shoe will always move first and will not let the other move.
