so my 2.7L tacoma has started misfiring. Ive gotten only one check engine light and that is when i totally lost cylinder four. i ordered new spark plugs which helped the problem but didn fix it. what is the next step in trying to solve this problem

did you replace the sparkplug wires as well? These get worn out too

Or if you have coil packs instead of the wires, swap the problematic cylinder's pack on another cylinder and see if the problem follows it. That's a quick way to test for a bad coil pack

I cannot guarantee it is what is causing the problem, but it definitely won't hurt to replace the sparkplug wires. It is actually recommended to replace them at the same time as sparkplugs

If the problem persists, you will at least have ruled out the wires

Are you sure yours has spark plug wires? My 2000 does not (its coil on plug).

If you can access the wiring connector to the four injectors, you might want to try disconnecting them and then measuring the resistance of each injector with an ohmmeter. Should read between 14 and 16 ohms.

    Sorry, I don't know how easy that would be on a 4-cylinder engine (I have a 6).
