I have a 2010 with the 2tr-fe, I'm tearing down to replace head gasket. I'm at the point of lining up timing marks before removing cams.

Do I have to roate it a lot of times before all marks on both chains line up?

The colored links on the chain are for installation purposes only, they will not line up once the engine is rotated.
Use the physical marks on the cam gears and crank gear (which should line up every 2 crankshaft rotations) to set the engine for chain removal.

    Touched other motors not 2.7.

always used manual referencing timing marks on crank and cams.
If I need to rotate them independently I do it with the valves closed so they don’t smack the piston.

I hope you have a helms manual or alldata etc… before tearing into this motor. Worth their weight in gold when you start doing engine work, believe me…
