I hate leak downs, never know if valves are really closed....
start with a cold compression test on your issue cylinder, & then just a known good cylinder.
if your problem child is much lower...........

    Plugs look decent
No obvious signs of coolant/HG issue

I really like what Slater suggested
Pull an injector and see if drips after shut down

Or could be a visual inspection

    Very recent thread…
Look at the red (old) compared to the blue(new)

F'k that guy, he's a dick & knows nothing about Toyotas, just a burned out ford monkey.....
    Seen it a few times...
If it is an injector, we dont know if its a bad spray pattern & restricted, or if its being hung-open after sitting & slowly dripping fuel into the cylinder....
On fords, they drip for often than restrict.
So just pulling them & cycling the key to pressurize the fuel system while monitoring the situation for a bit, you will get your answer....

Or even cranking the engine & watching the spray pattern....

But, I have no idea what it looks like under the hood so no idea how easy or difficult doing so is after you access them.

TD & I have been further discussing behind the scenes....
    Hey appreciates the knowledge...

    IMO
That’s why I get along/1st name basis with techs at most of my shops

I arrive at said shop and my finger nails are black/fresh cuts on hands….just like theirs…..of course some no bc they like to use those black sweat gloves(I don’t)
never asked but I’m sure techs notice that stuff

Most of the time when I've found a bad injector on a toyota it ran poorly at various times during a test drive, like the miss would show up and then go away.. show up, go away..

Just the fact that your miss is only at startup and then doesn't come back again until the truck sits and gets cold.. makes a leaking headgasket highly suspect.

Have you pressurized the cooling system and looked down in the cylinder for coolant yet? That's something that can be done before disassembling the intake manifold. It's relatively easy compared to digging injectors and the fuel rail out

I have a tree service taking down some trees in my yard so I haven’t had a chance to work on it.

After I changed out the spark plugs and wires now I have no misfire when starting the engine cold and as long as I don’t shut it off it runs fine no misfire.

When I drive it and bring it up to full temp shut it off and restart while full temp misfire is back. Let it sit all night start it up next morning runs smooth as glass.

    I hope this makes sense.

Can you get a stethoscope with a long metal probe back under there and touch each injector and listen? When the problem is happening?

The ones that I found to be bad had a different tapping sound than the rest. Like dull or muffled, not a nice sharp metallic tap, tap, tap..

Did you check the ignition coils closely for cracks or carbon tracking?

The injectors are under the lower intake but it might be possible and I do have a stethoscope so I will take a look at that tomorrow.

I changed out the ignition coil with no change.

Update: I had a little time this evening so I started the truck and as usual it ran fine cold. I got out my stethoscope and listen to the suspected bad injector and two others and they all three made the same identical clacking sound.

Then I took the truck around the block brought it up to full temp came back home shut it off waited 15 minutes started it up and it had the misfire on cylinder #2. I put the stethoscope on it and it definitely had a different clacking sound than the others. The working ones are making a clacking sound and #2 more of just a tick.

Update: it looks like I dodged the bullet. I moved the suspected faulty injector to from #2 to #4 and the misfire followed. I ordered an OEM injector this afternoon so looks like I got lucky. Thanks for all the replies!

    sweet,
Sounds like firing off an injector out of the parts cannon would have paid off here!
Now you get to do it twice...:D
