I push the button and it just flashes. I've followed the troubleshooting suggested in the owners manual without relief. The solenoid makes no noise when I push the button. I have taken the cover off and made sure the shaft inside the housing moves freely. Not sure if I should try and bypass the trucks wiring or how to go about legitimately fixing the issue. Suggestions and help appreciated.

If the solenoid isn't making any sound then it seems like a good idea to at least connect power to it to see if it's working. This may not be your problem at all, but another person had another issue with their e-locker and I guess it's just good to be read up on them in general if you have one:

Update: I removed and serviced the actuator as outlined in
    http://personalpages.tds.net/~thompson_wp/tacoma/locker.htm
with no resolution. The elocker switch has power and is functioning properly as the light was solid when the locker was engaged manually. I cleaned all wiring connectors and still have no power to the 5 pin connector on the actuator. 4wd fuse is good. Any ideas where to go from here?

When you switch it, it gets power to the motor? Could be a bad actuator. Check the motor for resistance.

I'm curious, I don't have an E locker, so how does it work?
Can you lock the rear diff at any speed? Does it work only up to a certain speed?
Do You have to be stopped to engage it??
How fast can you be driving with it engaged ?
Or is the E locker for 4 low speeds only?
    Or just crawling

The TRD e-locker is designed so that it only works in 4lo (if you have 4x4) or at speeds of under 15 mph if you have a 4x2 Prerunner. You don't have to be stopped to engage it, I've hit the button before in 4lo while moving and it usually engages after a few feet or so of driving.

If you do the 'grey wire' mod then you can engage the e-locker any time you want

If you engage while moving, do it while going straight and the wheels aren't spinning at different speeds.

The relay is in the locker ECU (PRERUNNER) or 4WD ECU 4X4

They're easy to open op and look for broken solder joints also.
