Try to keep your shift points below 2000 RPMS and drive as slow as you can on the highway. I drive 60 and get pretty good MPG, 65+ it gets worse and worse as you go up. If you drive 70 don't expect to hit 20 MPG. If you are in a hurry, one trip doing 85 MPH won't blow your wallet, but leave sooner so you can drive slower.

When I needed new tires I bought the Bridgestone Dueler Alenza Plus, improved my MPG by 2-3. They are a highway tire and I was honest with myself and knew I wasn't going to use an A/T anytime soon. If you go on any trails or offroading, stick with a lightweight A/T.

A bed cover will help but will take a couple years to pay off. No exhaust mod will payoff except for some members have reported better MPGs with replacing just the muffler with a Flowmaster 50.
Other than that, maintain the truck religiously with the best products

Bilstein 6112's 5th clip 1/4" spacer driver's side. Icon tubular UCA's with Delta joint, wheelers superbumps, ECGS bushing. Allpro standard leaf packs, 5160's, U bolt flip kit with wheelers super bump's, extended stainless steel brake lines, carrier bearing drop. Icon rebounds 17" with Nitto terra grappler G2's 285/70/17's.
I just checked my highway mpg's today. 94.5 mi, all highway. Filled up. 17.3 mpg. Came back the same dam road, to the same station I'd filled up earlier in the day. Filled up. 19.4 mpg. Go friggin figure!!!! Light winds, sunny day, terrain flatter than a pancake. Temps on the way down, 25-31°. Coming back, 31-36°. The (only) thing I can think of is the ecu adjusts rich-lean mixture @32°?

Bed covers don't help with mpgs. I think myth buster did a thing about this.
Here is a link to something similiar http://www.tundraheadquarters.com/blog/the-tonneau-cover-fuel-economy-myth/
Leave your tail gate up there is no reason to take it down it won't improve mpgs. Ride with your ac it won't make much of a difference to ride with your windows down besides being hotter.
