When I purchased my 2.7 L prerunner it came with green coolant inside. I have decided to replace it with the pink coolant designed for Toyota/Lexus vehicles. I want to flush all the old stuff out but I'm not sure if I should replace it with a 50/50 mixture or with concentrate (assuming there will be a bit of distilled water from the flush left in the engine). If I use just the 50/50 there will end up being more water in the mixture than there is supposed to be. If I have to use concentrate, how much water should I put in with it?

I think the pink coolant only comes in 50/50, but I could be wrong.

Ok, thanks for that info. Is it fine that there may be a bit more water than normal in the mixture?

You want it close to 50/50 as possible, but if it’s off a little it should be fine. You can by a cheap hydrometer at any parts store to check the protection temp level when you’re done, just to make sure you have enough coolant to water ratio.
