    I have a 2013 Tacoma Dual Cab TRD Off-Road.
Is it possible to use Differential Lock while in 4-High/ 4- Low?
Truck has 280,000 miles and didn't want to try it without asking
    the group first.

Stock, you can only use the rear locker in 4low. Use it (on a low traction surface), exercise it to keep the actuator operating properly.

There is a wiring mod you can do that enables you to lock the diff in 2wd or 4hi as well, with 1st gens it's referred to as the Grey Wire Mod. Not sure what the 2nd gens call it

Heres an alternative that i use for the locker anytime mod, way less invasive to your wiring harness

    https://www.toyotanation.com/threads/locker-mod-an-alternative.226409/

With the only caveat is that it will throw a check engine light if you go over 25mph (or travel x amount of distance). Easy price to pay for not having to chop everything etc just clear with a bluetooth OBD and a phone.
