I'm having similar issues with 16 Tacoma. I can't lock the doors from the driver side door but unlock works fine. I have to lock with the button on the front passenger door or with the key from the outside.

I probably should posted this after I found it but forgot too. This plug was lose, I pushed it back in and door lock buttons started to work again.

No..I think they're good ..door handle works and locks manually using the key from the fob . The sensor in the handle locks the other 3 doors but won't unlock them.:( this is driving me nuts .

Update ….problem solved .It was a broken wire inside the rubber boot that goes from the door to the body…. Small black wire with no slack in it, had to add a bit of wire to rejoin.
Thanks to AIbot01 and 099 for your help

In the hopes of helping someone in the future, I had the same door lock actuator failure happen on my 3rd gen. It actually was intermittent for years and eventually failed completely. I tried buying the small motor linked in this thread and replace just that piece but the motor did not match exactly what was in the actuator. The shaft was shorter and an oval shape.
The front door actuator complete oem part through Toyota is $370.
Instead I bought this knockoff on eBay for $40. So far it works perfectly, I will update if it ever fails. Don’t let Toyota rip you a new one.

Where do I find this rubber boot? So, just a quick recap of what I am experiencing: One night, my 2017 Taco started beeping three times loud, as if I had left a door open when locking it. I quickly found that my driver's door was the only one not locking with the key fob or the door button. So did some research and found I could have a bad actuator. Quickly called a local parts store with a buddy of mine and was quoted 225.00. Went on ebay and found a remanufactured unit for 80.00 plus a lifetime warranty with the core return. I went with the Ebay one. Replaced it in under an hour and it still did not work. I didn't realize there was fuse for the specific door so I checked it out. Fuse was good but I replaced it anyways just in case and still no cigar. I then got my multimeter and unplugged the plug that gives the actuator its power. I put my probes into the 2 upper pins and tested it and after testing the door locks with my key fob I am getting no current. I started thinking maybe I am testing this wrong...So I took apart my passenger door front door panel and ran the same test. I got about 12 to 13 volts on multimeter every time I locked or unlocked the door.

So now the problem still exists in my Driver side front door. So any suggestions would be helpful. I have a new Actuator and replaced fuses.

would I find this rubber boot?
AIbot01 where is that panel you posted in your picture? Is that by the brake pedal to the left?
