I have a 2tr-fe, I parked nose up on a steep hill wresting it in 1st gear as the e brake has stretched, can this rotate the engine backward and cause damage?

No, your engine is mounted to the frame. I'm not sure what you mean by rotate the engine, but the engine is not going to move out of place unless your motor mounts are broken or unattached.

I'm asking if it's possible to rotate the crank backwards and create slack in the timing chain causing the valves to hit piston

Bilstein/Icon 2" suspension lift with wheelers 5 leaf springs, 7-pin relocate through bumper, bfg KO2s, redline hood struts, immryo mirror relocation, stubby antenna, dipped emblem, heated mirrors, Jason trek cap, led fogs, debadged
If your timing chain has enough slack to make that a problem, you have bigger things to worry about.... Your engine should hold remarkably well in first gear.

Just that I heard somewhere that the tensioner can become slack as it is only meant to go one way

Leave it in reverse and it won't turn backwards, if it turns at all.

And get your Ebrake fixed ;)

Mine did roll away a few times while parked in 1st with no e brake. Didn't hurt anything that I could tell, although I was amazed that it actually rolled.

Did roll backwards in 1st and was it the old 3rz or the new 2tr-fe, not sure if the old engine was a interference type.

2RZ (so pretty close to the 3RZ), I believe it was an interference engine. Yes it was in 1st gear, it would roll on that specific driveway in 1st or reverse.

Mine still seems to be running ok, no noise on cold start up, but I will def leave in reverse next time and fix e-brake.
