I have a 2013 dcsb Tacoma, I'd like to know if it is better to run higher octane fuel or does it even matter. I'm having some clicking/ ticking noises at low speeds when accellerating and wondering if a higher octane fuel or a booster would help.

Higher octane fuel won't make a difference unless you're running some kind of forced induction (super/turbocharger). Does it sound like 'pinging' or pre-detonation when you accelerate? If that's the case, you should definitely look into it with a mechanic if you aren't able to yourself because these engines are designed to run on regular unleaded. If it 'needs' higher octane to run right then something's likely not right.

As mentioned above you should be able to run regular gas with no issues unless something is wrong. However one tank of fuel is a cheap experiment to see if YOU notice a difference. To the collective feel free to correct me if I am wrong but the computer has a means through sensors to advance and retard spark on the engine and should be adjusting for octane level?

This is completely normal for the 4.0. Sounds terrible I know, but nothing to be worried about. I fixed it with a supercharger, now I just hear the whine. :)

Unless your supercharger causes pinging which mine does from time to time :( the 3.4l setup is notorious for it, I really need to get the 7th injector kit...
