To be quite honest I don't think you are going to get the difference you are looking for , and will probably spend more in conversion to the JBL than just doing aftermarket. I would do some consulting with Mr. Marv or pose the question in the A/V section.

some of this, a little of that
If I were going to spend the money I think I would go with some good aftermarket speakers and a nice head unit. The JBl stuff is nice but not nearly as good as you can get aftermarket. I just don't think you would get the performance gains from the JBL you are looking for. I think the JBL sounds better than stock but it can't compare to a good set of aftermarket speakers and head unit.

So this thread appeared to die without ever getting answered. I would like to keep my '05 Tacoma as stock looking as possible on the inside and have found a complete JBL system for relatively inexpensive on craigslist. Have searched everywhere but can't find out if it is possible to upgrade the factory single-disc non-JBL stereo to the 6 disc JBL unit. Anyone know? Thanks.

One connector is different, but I could not find it anywhere. I bought the JBL off ebay because I like the stock look too and a miner upgrade.
I think it comes down to finding a wreck and getting the stock toyota Wiring harness.
