When I hit the lock button on the driver door, the driver door does not lock (the passenger door does lock). Any advice?
I can manually lock the driver door with the key or by pushing the lever (the part with the orange sticker showing when it is unlocked).
    Any help is appreciated!

There are settings from the nav screen that enables you to choose which doors lock and unlock when you lock the doors. Not sure if it's for the key fob or door handle only, but you can give it a shot.

    I don’t have a nav screen.
Sometimes the door locks. Sometimes it doesn’t.

Sounds like your door latch is bad (the electrical part of it).
We see a lot of these going bad in the Lexus world.

Pull apart door, remove solenoid/latch assembly.. clean with degreaser, use a good lithium grease spray to lube.. reinstall and check function.. if it fails to unlock/lock again, replace the latch assembly. should take no longer than 30 minutes to get it out.. 30 min to re-install. solenoid may just have become weak, or the mechanism inside it may be severely worn..
