I just temporarily installed a set of 2" tapered blocks with some 5100 billies. Which side faces back. The bigger end or the smaller end. I put them in with the bigger end facing forward but not sure if this is correct.

Bigger end faces back. The angle should help tilt the input flange in the axle upward, toward the carrier bearing.

So being that I put them in backwards. Would this be contributing to my vibration at around 30 mph.

Absolutely. Your universal joints won't be happy if you put the blocks in backwards.
