I've noticed these little gray plastic brackets on the frame. See yellow arrow pointing to them in the picture below. What are they? I've seen pictures of the frame in a similar area, relating to it being a jack point, but the frame didn't have them. Thanks!

Drain cover to let the frame drain without water splashing up into it. You should have another one on the other side.
If you put a jack under it you’ll break it, it’s just plastic.

They are factory on all 3rd gens. Most get busted by jacks/jack stands... As another poster suggested, it's been debated before and most agree it's to block a drain hole in the boxed portion of the frame.

Just took them out of my frame when I was putting it on the lift the other day. Was wondering what they were as well

Stock wheels and suspension. Random "utilitarian" mods featured on this great forum.
They're definitely there on my '20. At first, wondered if they were leftovers from assembly but yeah, drain cover sounds a lot more believable.

That makes sense. Thanks for the info. Does it matter which way they are facing? The one on the passenger side is oriented 90 deg off the one on the driver's side... at least on my truck. They have arrows printed on them, but I'm not sure what the arrows would be referencing.

I’m not sure what the arrows mean but both of mine are pointed to the front/rear. DE66D8D4-299B-4C54-979A-CFCFEFDD3F53.jpg87DE3E3F-208B-416F-8C83-7A2FB6FF8DA6.jpg

Any negatives to not having these installed?

I would of noticed them when i sprayed the frame for rust proofing, and later inspecting it a year later. They either got hacked, off or were never installed. I've got several of these rubber frame plugs though.

Yes. They protect the sensors that put the truck into submarine mode when they contact water.

Those parts are more complex than they look. The part is a splash guard for the drain hole as others have mentioned, but it is also a “snorkel” that prevents the drain from being clogged by whatever silt accumulates in the bottom of the boxed section. It has multiple stacked entry holes, up inside the boxed section, so if the box is filled with silt, or that white CRC anti-corrosion snot that Toyota is injecting in there under the recall, water sitting on top will not be trapped in the box and will drain out the snorkel.

I’ve broken a couple and they are like $2 at the dealer.
