    Consider trying something simple.

You say this is a 2010 truck with the 5 speed tranny and V-6.

Sounds to me like the tranny isn't shifting into 4th or finding OD.

Do you know when the tranny fluid was changed? Is the fluid level low? Miles on the truck?

You could try a fluid/filter change for the tranny. Pretty simple task, just a little messy. Parts at RockAuto for <$15. Fluid will be the most expensive items of this service.

Low or dirty or worn out fluid will cause shifting problems.

I too think this sounds like a stuck solenoid. Esp since it has been sitting for extended periods of time as per the OP.

    Yup

I am going with an electrical issue as well .
Chances are the check engine light will not be illuminated for this one .
Considering this is a completely computerized transmission ,It will have to be scanned for codes .

Some things that prevent overdrive and the converter clutch from functioning properly are the brake switch , transmission temperature , throttle position sensor , neutral safety switch ,brake switch , 4th gear switch and solenoid control .
Without scanning for codes , we are just guessing.

The issue fixed itself? Didn't drive it for a couple days. Today, 85 mph on the highway-2100 rpm? Wtf? Waited a half hour did it again. Same. Tried some accelerated starts and nothing like I had seen that day. Maybe the tire place put the vehicle on some machine to balance or align? Maybe just needed a cool of afterward? I don't know, but still getting it looked at.
It's at about 106000 mi. Maintained to factory spec from day 1. Always on time with oil changes and recommended maint. from Toyota. But that was just weird. Sorry to get everyone involved in a non-issue. And thanks for the responses.

I've heard bad things about getting them looked at without the symptom happening. Glad it's not happening, maybe something in the computer system had to reset or something similar.

Stuck solenoids will sometime get freed up by themselves... Hope the issue don't return!

Welcome to TW. As for the tailgate, climb in the bed and see if you can remove the screws to remove the plastic panel. You should be able to see the issue with it with the panel off while lifting the tailgate handle. Likely the connecting rod from the locked side came loose. You can manually unlock that side by manually pulling on that rod. Once you are able to repair it, I suggest spraying all connecting components of the handle assembly with lubricant.

Wattapunk, I'm on it. Just a matter of wanting to work out in the cold. No room in garage.

Fairly easy fix. You can see where the rod hooks onto the swivel component. Yours likely came off. Just for reference with the panel off:
