Awesome CB setup, TRD leaf springs, matching camper shell...more to come some day.
Head gasket or leaky injector would be my suspension...lots of fuel in a cylinder at start up will cause a misfire till it clears. A head gasket that is starting to go might start to swell after it starts up and give you compression back?

Obviously I'm just spit balling....maybe a bad coil or something else simple as well.

How many miles do you have on the plugs?

You need to find out for sure if it is a leaking injector. If it leaks enough it will hydrolock that cylinder when you start it and bend a rod or break a piston.

Well see it has a new fuel injector from factory and new spark plugs from dealer. Coils I swapped them from another cylinder and still does the same, BUT hers the catch and I know if it has to do something with it. I notice I was low 1 qt of oil just when I was about to do its oil change

Is it every start up, or primarily on cold start up?
Maybe you have a head gasket leaking internally on a cold engine?
Does your coolant level drop any at all?

Maybe pull the plug before the next cold start and look into the cylinder.
You may need an inspection camera to see fully into the cylinder.

a weak ignition coil wire to that cylinder

    a worn spark plug

true it could be an injector or gasket

check from that cylinder back............... easier & cheaper

"a new fuel injector from factory and new spark plugs from dealer."--unquote. Check the spark plug on that single new injector cylinder,,lets see what you see. Why just one injector on that visit?,,why not a matched set like they should be?. I would be wary of a toss-in? injector. Can cause a real tail chase until you circle back,,again. Hmm. Connectors on the recently wrestled injector harness?. Check the plug wires for resistance,,there is a value per foot of length on plug wires and they were recently wrestled as well. Make sure they snap/click real nice going on, , and as well use dielectric grease on both ends of the plug wires. Eliminating the nit picky stuff take time and patience.

Try swapping #2 plug with another. Grab codes again.

If you suspect a leaky injector you could stick a LONG cotton swab down there until it hits the piston, and see if it comes up wet or at least smells strongly like fuel. Do the same with the other cylinders to compare. (EDIT: Do this BEFORE you start the engine after it's been sitting, obviously.)

You can also yank the injector harnesses individually as it runs,,see if one individually really rattles the engine mounts. Don't do this for a long period,,just a quick check. Wear rubber chemical gloves. It Might be your problem child. It's gonna throw codes due to that. If you have a code reader and resetter info,,no big deal then. Done it a few times over the years on my 1st gen 3RZ with no issues, Just to make sure everyone is playing nice and even at idle. A timing light with rpm indicator,,to watch the drop per cylinder underhood.
