I replaced my rear diff oil today ('98 Tacoma base model 2.4L) and the amount it took to reach the bottom of the fill hole was noticeably more than what the OEM manual says is required (the Chilton manual had the same number): 1350mL.

I bought two 946mL bottles of Mobil 1 75W-90 (1892mL total) and I have about 215mL left, which means I poured in about 1677mL of oil.

So, does that mean the extra 327mL to get all the way to the bottom of the fill hole isn't really necessary?

You've way overthought this. Don't measure in milliliters and just fill it up til it pours out the fill hole.
