I have sprayed PB Blaster in the lock cylinder from the outside and as best I could from the inside. The key can manually lock the door fine, but the cylinder fails to turn when attempting to lock.

It’s either a worn key, a bad hardware store copy or a problem with the lock cylinder. There’s a black plastic plug and 1 bolt in the edge of the door. That’s all it takes to remove the lock. Take your registration and ID to the dealer and ask for the key code number for your truck. Take that number to a locksmith. Then let the locksmith determine if it’s a key problem or a lock problem.

The key had worked for years. It still works fine in the drivers side door, ignition, and locking the passenger door. My thought was something mechanical is wrong with the lock cylinder. Thanks for the information though I'm getting that cylinder out as we speak.

It could still be a key problem. The key wears and the locks you use the most wear so they work. The passenger side lock doesn’t get used much so it requires a more precise key.
